Amakhosi reached the semi-finals of this tournament last season but lost out on reaching the final to arch-rivals Orlando Pirates after a 2-1 loss in extra-time.
The Glamour Boys will be without left-back Sifiso Hlanti due to suspension.
Milford, meanwhile, are winless since re-opening their Motsepe Foundation Championship season after a 1-0 loss to Orbit College and a goalless draw against Maritzburg United last weekend.
The KwaZulu-Natal side are lying 12th on the log, just a point above the relegation zone.
The Stallions were in the last 32 in 2016 but lost 2-1 to neighbors Golden Arrows.
