This comes after the 30-year-old returned to Stamford Bridge in July following the expiration of his loan spell at Inter Milan.
Lukaku, however, was not considered part of Mauricio Pochettino's plans in London, while prospective permanent moves to Inter and Juventus fell through for various reasons.
Speaking after his switch was made official, the forward said he has been thrilled by the way he has been welcomed at his new club.
"The welcome I've received from this club and its fans has excited me and given me even more motivation to give my all for my new team," he told the Roma's website.
"As an opponent, I felt the atmosphere of the Stadio Olimpico, and felt the warmth of the Romanisti.
"It's nice to know I'm a part of it today. I recently got the opportunity to speak with the owners, and I was impressed by their ambition. Now we must work, be humble, and grow game by game. For my part, I can't wait to be available to my teammates both on and off the pitch."
The left-footed forward contributed to 21 goals in 37 matches for the Nerazzurri last season, and will now look to help Mourinho's efforts in Rome.

Lukaku has played for Mourinho twice before, at Chelsea and then Manchester United, scoring 33 goals in 76 appearances overall under the tactician.
