Sundowns booked their spot in the semi-finals after beating University of Pretoria on penalties following a 1-1 draw in extra-time.
The Brazilians were crowned DStv Premiership champions for the seventh consecutive season after trouncing ten-man Kaizer Chiefs 5-1 courtesy of a brace by Tashreeq Matthews and goals by Matias Esquivel, Lucas Ribeiro Costa and Thembinkosi Lorch.
Stellenbosch, meanwhile, secured a semi-final berth after a resounding 4-0 win over SuperSport United in the quarter-finals, thanks to goals by Anicet Oura, Devon Titus, Andre de Jong and Iqraam Rayners.
Stellies maintained their second position in the DStv Premiership table after thrashing Golden Arrows 3-0 on Wednesday, with Titus, Oura and Rayners on target.
The last time the sides met in the Nedbank Cup, Steve Barker's charges eliminated Masandawana 2-1 in the quarter-finals in April last year.
