The Brazilians have had a quick turnaround from last season and only started pre-season a couple weeks ago following their involvement in the FIFA Club World Cup last month.
Sundowns will be hoping to improve on last season's showing in the tournament after losing to Stellenbosch FC over two legs in the semi-finals.
Miguel Cardoso's charges have only brought in one new player during the transfer window following the arrival of Bafana Bafana defender Fawaaz Basadien.
Richards Bay, meanwhile, are set to make debut appearance in the tournament after securing an eighth-place finish last season.
The Natal Rich Boyz were involved in the KwaZulu-Natal Premier's Cup last weekend, where they reached the final and beat Durban City to secure the title.
