Brandon Truter's charges made it back-to-back wins after defeating Kaizer Chiefs 1-0 at the FNB Stadium seven days ago.
The two successive victories have pushed Sekhukhune up to 12th position, four points adrift of the relegation zone.
Stellenbosch, meanwhile, extended their winless run to seven matches in the DStv Premiership after drawing 1-1 with Marumo Gallants on Wednesday.
The draw has left Steve Barker's troops one spot and one point behind Sekhukhune heading into this evening's encounter.
