Both legs of the second preliminary round tie will be played at Loftus Versefld, with the return fixture taking place on Friday.
The Brazilians were given a bye in the first round, but have been keeping busy thanks to their domestic schedule with their last encounter ending in a goalless draw against Orlando Pirates last weekend in the MTN8.
Sundowns will be hoping to improve on last season's showing in Africa's premier club competition after reaching the quarter-final stage, where they were knocked out by Petro de Luanda.
The Brazilians will come up against La Passe, who overcome Comoros-based side Volcan Club in the previous round.
This will be La Passe's first encounter against South African opposition, although Sundowns have faced opposition from the Seychelles as they defeated Saint Michel 4-1 on aggregate in the 2015 preliminary round.
