With Chiefs sitting in seventh spot in the Absa Premiership, the Nedbank Cup looks to be the last chance for the club to end a four-year drought without a trophy.
Ernst Middendorp's charges faced ABC Motsepe League sides in the last 32 and the round 16, and on both occasions they were taken to extra-time.
Meanwhile, the Citizens had tough opposition en route to the quarter-finals as they beat SuperSport United in the round of 32 before seeing off Highlands Park 3-0 in the previous round.
Benni McCarthy's troops are still hungry for success after winning the MTN8 Cup earlier in the season and the Nedbank Cup remains the only knockout trophy they have not won since they formed in 2016.
The sides have already met twice in the league this season as Amakhosi claimed a 4-1 in the first round in Cape Town before the Citizens won the return game 1-0 at FNB Stadium.
