The defending champions, who won the trophy in 2008 and 2012, had left a few big names – including Juan Mata, Diego Costa and Fernando Torres – out of their preliminary squad.
Now, Isco and Saul have been scratched from Del Bosque's selection as he had to cut the team down to 23 names ahead of the tournament.
Meanwhile, Arsenal's Hector Bellerin, who wasn't in the preliminary squad, was called up to replace the injured Dani Carvajal.
Spain have been drawn in Group D alongside Turkey, Croatia and Czech Republic.
