Pirates won the MTN8 back-to-back after beating Mamelodi Sundowns on penalties following a goalless draw in extra-time.
The Buccaneers have reinforced their squad with the likes of Angolan Gilberto, Selaelo Rasebotja and Thuso Moleleki.
The Soweto giants will be without Tapelo Xoki and Paseka Mako who are still nursing injuries.
SuperSport, on the other hand, were knocked out in the first round last season after a narrow 1-0 defeat to Stellenbosch in extra-time.

Former Orlando Pirates winger Vincent Pule is among Matsatsantsa's new signings, that also includes Pogiso Sanoka, Nyiko Mobbie, Brooklyn Poggenpoel, goalkeeper Thakasani Mbanjwa and Keanin Ayer.
The sides have not met in a cup tournament since 2018 when SuperSport knocked Pirates out of the MTN8 in the quarter-finals on penalties after a 2-2 draw in extra-time.
United came from behind as goals by Dean Furman and Evans Rusike put them in the lead after Augustine Mulenga had given the Buccaneers an early lead.
Gladwin Shitolo forced found the equaliser for Pirates on the hour-mark.
