After a losing start to the season, with a 1-0 defeat to Stellenbosch, Pirates redeemed themselves on Tuesday after thrashing Royal AM 4-2.
The Buccaneers were crowned champions of this competition for a record four times under current sponsors and kept the trophy for good after edging AmaZulu 1-0 in the final last season.
Sekhukhune, meanwhile, are participating in this tournament for the first time after finishing seventh on the log last season.
Babina Noko also had a losing start to the season after going down 2-1 against Mamelodi Sundowns but they redeemed themselves with a 2-0 victory over Cape Town City on Wednesday.
Pirates beat Sekhukhune 2-1 in the Nedbank Cup final last season but they could not beat Babina Noko in the league, as they lost to them 2-0 and drew with them 1-1 in the reverse fixture.
