Ajax Cape Town and Tshakhuma Tsha Madzivhandila (TTM) kick-start the Promotion-Relegation play-offs on Sunday afternoon at Rand Stadium.
The Urban Warriors have another chance of being promoted to the top flight after narrowly losing out on automatic promotion to Swallows FC by goal difference last weekend.
TTM on the other hand, qualified for the play-offs after finishing third in the GladAfrica Championship and had already sealed their place before the final game of the season and lost 3-0 to Swallows FC.
TTM will be campaigning in the elite league next season after purchasing Bidvest Wits' top-flight status and sold their National First Division status to fellow Limpopo side Sekhukhune FC.
The two sides will be joined by Black Leopards, who finished 15th in the Absa Premiership, in the play-offs that will contested in a mini-log format.
Vhadau Vha Damani recently beat Ajax 1-0 in their second match in the bio-bubble after the Cape Town side won the first-round clash 3-2.
