Matsatsantsa have had a tough start to the second round of the campaign after back-to-back defeats against Maritzburg United and Royal AM.
Kaitano Tembo's charges have since dropped out of the top eight, lying ninth on the table with 26 points and need a win to get back into the top half of the standings.
Usuthu, meanwhile, are winless since the restart, after playing to two draws against TS Galaxy and Stellenbosch. However, Benni McCarthy's men are doing well in the CAF Champions League, having picked up two consecutive wins against Horoya and ES Setif.
On the domestic front, AmaZulu are placed sixth on the table with 29 points and will be eager to climb up the ladder and compete for continental positions.
The sides could not be seperated in the first round meeting after playing to a goalless stalemate in September last year at Jonsson Kings Park Stadium.
