United manager Erik ten Hag gave a Premier League debut to Willy Kambwala in place of Raphael Varane, who missed out through illness while Marcus Rashford started on the bench once again.
The Red Devils dominated proceedings in the early exchanges of the first half but they were unable to find a way past the Hammers defence, who were sat quite deep in their own half.
United did get the first big chance of the game after 35 minutes when Antony sent Alejandro Garnacho through on goal, but the young Argentine could only fire straight at Alphonse Areola.
Much like their goalless draw against Liverpool last weekend, the first half failed to provide any more big moments, much to the annoyance of the West Ham supporters before the halftime whistle.
Ten minutes after the break, West Ham got their first chance from a corner when Jarrod Bowen got on the end of James Ward-Prowse's corner but saw his header tipped over the by goalkeeper Andre Onana.
Bowen was not to be denied in the 71st minute when the English combined with Lucas Paqueta before bundling the ball past Onana for his first-ever goal against the Red Devils.
The lead was doubled in the 78th minute when Kobbie Mainoo lost possession and gifted the ball to Paqueta, who in turn played in Mohammed Kudus to carve out some space before firing past Onana.
The Red Devils were unable to find a way back in the contest in the latter stages of the second half, as they slumped to an eighth defeat of the league campaign.
