Coach Ruben Amorim brought the regulars back into the team after the League Cup exit, with Bruno Fernandes, Bryan Mbeumo, Casemiro and Mason Mount getting recalled while Bafana Bafana striker Lyle Foster was tasked with leading the Burnley attack.
The Red Devils made a lively start to the contest and were awarded a penalty on 15 minutes when Kyle Walker fouled Mount in the box. However, after a review on the Video Assistant Referee (VAR) monitor, the referee overturned the decision.
The hosts got their reward in the 27th minute when Burnley's Josh Cullen inadvertently deflected the ball into his own net after Casemiro's header had been kept out goalkeeper Matt Dubravka.
United were dealt a big blow two minutes later when the lively Matheus Cunha was forced off with an injury and replaced by Joshua Zirkzee, who was making his first league appearance of the campaign.
The hosts should have doubled their lead advantage on the stroke of halftime when a swift counter-attack ended with Bryan Mbeumo teeing up Amad Diallo, who blazed over the crossbar with only Dubravka to beat.
Burnley, however, hit back ten minutes after the second half as Foster got his first goal of the season by turning home Burn Larsen's superb cross to open his account for the season.
The Red Devils hit back straight away when Diogo Dalot broke free on the left flank from Zirkzee's header before picking out Mbuemo, who makes no mistake from close range.
It was United's chance to switch off at the back as Burnley levelled matters for a second time on 67 minutes when Anthony bundled the ball home after the defence failed to deal with a corner.
Coach Ruben Amorim threw on Benjamin Sesko for the final 15 minutes and he nearly restored United's lead in the final ten minutes with a towering header that sailed just over the crossbar.
United were then awarded a penalty stoppage time after Amad was brought down in the box following a VAR check. Fernandes, who missed against Fulham last week, stepped up and calmly slotted home his spot-kick to seal the first win of the season.
