The Frenchman won the award ahead of the South African in second place and Venezuela's Deyna Castellanos in third, and was delighted to receive the accolade for his scorpion kick goal against Crystal Palace in January.
"First of all, I have to say it's an honour for me," he said, as quoted by the Evening Standard.
"I'm delighted to receive this trophy today in front of legends of football. Now l'd just like to thank the people who voted for me. I would like to congratulate the 10 nominees who scored amazing goals too.
"Obviously, I would like to thank my teammates, without them I couldn't score maybe this goal, and my family."
Giroud picked up 36.17% of the overall votes, while Masuluke received only 27.48%. Overall, 792 062 fans voted for their favourite goal on FIFA's official website.

Despite losing out to Giroud, Masuluke still made history by becoming the first African to finish among the top three in the Puskas Award ranking.
With disgruntled fans having referred to the prize as a "popularity contest", do you think the Puskas Award should be decided by a panel of experts instead of online voting by fans? Have your say in the comments section below.
