Chiefs got back to winning ways in the league after a late 1-0 win over Sekhukhune United thanks to Mfundo Vilakazi last weekend, which moved them up to fifth on the table.
The Glamour Boys were knocked out at this stage last season by Motsepe Foundation Championship side Milford on penalties after a goalless draw in extra-time.
Free Agents, meanwhile, will be playing for the first time in the tournament proper, coming from the Gauteng ABC Motsepe League.
Olaitan Oladeru's charges are lying 11th in the standings of 18 teams and will be aiming to cause an upset.
Chiefs will still be without head coach Nasreddine Nabi on the bench, who will be serving his second game suspension.
