Miguel Cardoso's charges bagged their first win in the league last week after defeating AmaZulu 2-0 at Loftus Versfeld Stadium.
Sundowns were involved in MTN8 action over the weekend after playing to a 1-1 draw against rivals Orlando Pirates at Orlando Stadium.
Cardoso will be hoping to brush Magesi aside as they have the second leg against Pirates coming up this Saturday.
Magesi, meanwhile, are searching for their first win after starting the campaign with back-to-back draws against Polokwane City and Stellenbosch FC.
