The 24-year-old was omitted from Bafana Bafana's Africa Cup of Nations squad but resumed domestic matters on Friday night with Ajax, after returning from their mid-season camp in Qatar.
Serero started and lasted the full 90 minutes in the middle of the park for Frank de Boer's side, as a solitary strike from Ricardo Kishna and an own goal handed the defending league champions the three points.
The win has moved Ajax to within one point behind league leaders, PSV Eindhoven, who are in action on Saturday.
