The Chilli Boys came close to lifting the trophy last year after reaching the final but narrowly lost 1-0 to Tshakhuma Tsha Madzivhandila (now known as Marumo Gallants).
Chippa have steadied the ship since appointing Kurt Lentjies as caretaker coach, as the Gqeberha-based side were on a four-match unbeaten run in the league before the recess last year.
The Chilli Boys have bolstered their squad with the signing of Ronald Pfumbidzai and Siphelele Luthuli both from Royal AM.
AmaTuks were in and around the promotion play-off spots in the GladAfrica Championship at the start of the season but have since dropped down to eighth on the log.
The Tshwane side were knocked out in the first round of this tournament last year after a 2-1 defeat to city rivals JDR Stars.
AmaTuks' best finish in the tournament was reaching the final in 2009 but lost 1-0 to Swallows, while they will face Chippa for the first time since 2016.
