After missing out at the weekend, Tau was handed start for Brugge while United's five changes saw Sergio Romero, Eric Bailly, Scott McTominay, Juan Mata and Odion Ighalo starting.
United made a strong start and hit the post with a well-struck shot from Bruno Fernandes following a quick counterattack three minutes from kickoff.
Brugge were down to ten-man after Ivorian Simon Deli was caught handling McTominay's curler to concede a penalty following a lengthy check by the Video Assistant Referee.
It was left to none other than January transfer window signing Fernandes who calmly send Mignolet the wrong way 27 minutes gone.
Ighalo got his United career off the mark after 34 minutes when a cut back from Mata down the right side found the Nigerian, who needed no second invitation.
The Red Devils upped the ante and netted a third in the last four minutes of the half through McTominay, who applied a top-drawer finish following a layoff from Fred.
United started the second half with the game's first change as Daniel James made way for teen sensation Tahit Chong.
The Red Devils remained ruthless and looked for a fourth after Mason Greenwood's shot from a distance shaved he post in the final 20 minutes.
Good work down the left from Jesse Lingard on 82 minutes did ensure their fourth as a low cross met an unmarked Fred who tapped home easily.
United still went out for more and Fred grabbed a double with a first-time finish from the edge of the area in added time, given the hosts a comfortable last 16 passage.
