While the Blaugrana have already qualified for the last 16, the visitors need to match Inter Milan's result against PSV Eindhoven in order to progress to the knockout stage.
Tottenham lost the reverse fixture at Wembley 4-2 but have since picked up seven points and enter the final group stage round in second position, level on points with Inter but ahead of the Italian side courtesy of their superior head-to-head away goals tally.
Spurs have never beaten the Spanish side, picking up one draw and two defeats in previous matches against the Catalan club.
If Barcelona, who became the first side to finish the group stage as leaders for a 20th time after securing the top spot on matchday five, avoid defeat to their English opposition, it would be the 12th time they go undefeated through the group stage.
Barca are undefeated in their past 28 UCL home matches, a run that began in September 2013, winning 26 of those games and drawing two. If they do not lose to Spurs tonight, they will equal Bayern Munich's competition record of going 29 home games unbeaten.
The Lilywhites, meanwhile, have won just four of their past 17 European away matches, drawing six and losing seven.
