Pep Guardiola made a number of changes with one eye on the UEFA Champions League this week, as Ederson, Jeremy Doku, Julian Alvarez, Manuel Akanji, Rico Lewis and Matheus Nunes came into the side.
The Citizens got off to the perfect start when a scramble in the box ended with Erling Haaland's shot taking a big deflection off and sailing into the back of the net.
It was one-way traffic from thereafter and they nearly got the second goal in the 27th minute when Kevin de Bruyne played in Nunes, who turned well before seeing his low strike crash against the upright.
That proved to be the trend for the remainder of the first half as City were camped in the opposition half, while Luton failed to carve out a single chance at Ederson's goal in the opening 45 minutes.
City eventually got their second goal just after the hour mark as Mateo Kovacic sent a superb volley into the top corner after getting on the end of Alvarez's well-weighted cross.
The home side went on to earn themselves a penalty in the 75th minute when Doku was brought down by Wilfred Onyedinma. Haaland stepped up and calmly slotted home his 20th league goal of the season.
However, Luton did pull a goal back five minutes later when Ross Barkley won the ball high up and skipped past two challenges before burying the ball into the far corner.
City did restore the three-goal advantage a few minutes later as Doku rounded off the scoring with a composed finish into the far corner after weaving his way into the box.
There was enough time for City to add a fifth goal in second half stoppage time when Joško Gvardiol collected a pass from Doku before rifling the ball home from the edge of the box.
