Juan Mata was given a rare start in possibly his last game for the club at Old Trafford along with Nemanja Matic who will be leaving at the end of the season.
The visitors looked lively in the early stages but it was United who broke the deadlock on nine minutes when Anthony Elanga got behind the Brentford defence and picked out Bruno Fernandes who volleyed home from close-range.
Brentford remained lively whenever they were in possession of the ball and went close to pulling level on 19 minutes when Christian Eriksen picked out Ivan Toney, who headed over the crossbar.
It was a fairly open contest in the first half as Mata went close to doubling United's lead with a snapshot on 33 minutes before Bryan Mbeumo fired into the side netting at the other end.
United had the ball in the back of the net just before the break when Ronaldo turned home Mata's cross, however the Portuguese was given offside by the Video Assistant Referee.
Fernandes had the chance to double the home side's lead seven minutes after the break after being slipped through on goal, but the midfielder dragged his strike wide of the mark.
De Gea was called upon at the other a few minutes later, with the Spanish shot-stopper deny Toney with his foot after United failed to clear their lines.
The Red Devils were then awarded a penalty on the hour mark when Ronaldo had brought down in the area by Rico Henry. The Portuguese dusted himself off and made no mistake to double his side's lead.
The home side managed to bag a third goal after 72 minutes as Raphael Varane got his first goal for the club with a volley straight from Alex Telles' corner.
Both Matic and Mata were replaced for the final 15 minutes and received a big applause in what was their final game at Old Trafford, with Phil Jones and Edinson Cavani brought on to see out the remaining stages of the win.
Here are 12 United players who could reportedly leave the club at the end of the season:
