Ashley Young and Scott McTominay made a return to the Red Devils starting line-up in place of Brandon Williams and Andreas Pereira, while the injured Anthony Martial was replaced by Mason Greenwood.
The hosts got off to a perfect as Marcus Rashford latched onto a loose ball before firing home at Paulo Gazzaniga's near post from the left side of the box.
Rashford was certainly in the mood and he was unlucky not to have bagged a second goal on 24 minutes when his effort from distance crashed against crossbar.
After slow start to the half, Spurs restored parity with their first real effort on 39 minutes as the in-form Dele Alli flicked the ball past two defenders before firing past David de Gea.
The Red Devils were afforded a chance to restore their from the penalty spot two minutes after the break following a clumsy foul from Moussa Sissoko, which allowed Rashford to slot home from 12-yards to put his side ahead.
Mourinho was the first to re-shuffle his pack just after the hour mark as Harry Winks made way for Danish midfielder Christian Eriksen, who has dropped down the pecking order this season.
With Spurs chasing the game, Ole Gunnar Solskjaer then made his first substitution with 11 minutes remaining as Pereira replaced Greenwood for more numbers in the middle of the park.
In the end, United were able to see out the closing stages of the second half to claim a vital three points in their pursuit of the top four.
