Setien replaced Ernesto Valverde at the start of the week after the former Barça boss was sacked in the wake of a 3-2 loss to Atletico Madrid in the Spanish Super Cup semi-final.
Blaugrana’s last league outing was a 2-2 draw with Espanyol in their opening game of the new year, allowing Real Madrid back in the hunt.
However, Zinedine Zidane’s charges moved three points clear at the summit on Saturday after edging Sevilla 2-1, but Barca will reclaim top spot with a win.
Blaugrana will be without frontman Luis Suarez who underwent knee surgery while Frenkie de Jong is suspended after being red-carded in the draw with Espanyol.
Meanwhile, Granada were last in action last Saturday after edging fourth tier side Temaraceite 1-0 in the second round of the Copa del Rey.
El Grana’s last outing in LaLiga ended in a 1-0 victory over Mallorca at the start of the year which bumped the side up to 10th spot.
The Catalan giants will also be looking for revenge, as Granada beat Barça 2-0 in the reverse fixture earlier this season for the first time since April 2014.
Here's how they line up:
Barcelona: Stegen, Sergi Roberto, Piqué, Umtiti, Alba, Busquets, Rakitic, Vidal, Griezmann, Messi, Ansu Fati.
Granada: Rui Silva, Víctor Díaz, Germán, Duarte, Foulquier, Gonalons, Eteki, Azeez, Vadillo, Machís, Carlos Fernández.
