Carlo Ancelotti opted to rest the likes of Karim Benzema, Toni Kroos and Vinicius Junior ahead of Wednesday's UEFA Champions League semi-final.
As a result, Mario Diaz was given a rare start in the attack while Eduardo Camavinga and Dani Ceballos came into the midfield for Los Blancos.
The home side nearly got the opening goal after 13 minutes when Casemiro headed the ball back across goal for Mariano who saw his effort tipped onto the past by goalkeeper Diego Lopez.
However, Los Blancos got their reward on 33 minutes when Rodrygo turned provider to pick out his countryman Marcelo, who slotted into the far corner from the edge of the box.
The lead was doubled two minutes before the halftime break through Rodrygo, who pounced on a loose ball before going through on goal to slot past Lopez.
While Espanyol struggled to really trouble Thibaut Courtois in the Real goal, the hosts went on to bag a third goal 10 minutes into the second half thanks to Asensio's finish.
With the hosts comfortably leading, Ancelotti introduced Benzema and Isco for the 30 minutes while Espanyol threw on Manu Morlanes and Wu Lei.
The substitutions made the desired effect for Los Blancos as Benzema turned provider to set up Isco, but the strike was disallowed for offside.
Benzema has been the standout performer for Real this season, so it was no surprise that he rounded off the scoring in the 81st minute with his 26th league goal of the season.
