Ruben Dias missed out through injury and was replaced by Fernandinho at centre-back while Bernardo Silva and Phil Foden came in for Jack Grealish and Gabriel Jesus.
Pep Guardiola's charges broke the deadlock after just seven minutes when Kevin de Bruyne latched onto a through-ball from Silva before sending first-time strike into the far corner.
Wolves, however, were back on level terms in the 11th minute when a swift counter-attack ended with Pedro Neto teeing up Leander Dendoncker to slot past Ederson from close range.
The hosts joy didn't last too long as City were back in front five minutes later, with De Bruyne bagging his second goal with another left-footed finish in the box.
De Bruyne capped off a lively opening 25 minutes by completing his hat-trick with a stunning curling effort from the edge of the box that left Jose Sa with no chance.
Wolves came out firing in the second half in search of a way back, but it was City who got the next goal as De Bruyne got his fourth with a right-footed finish this time.
With City comfortably leading, Guardiola opted to withdraw Fernandinho and Foden for the final 15 minutes as Riyad Mahrez and Grealish came on to see out the closing stages of the win.
The league leaders would go on to get a fifth goal but it wasn't De Bruyne, as Joao Cancelo latched on to Grealish's pass before his deflected cross found Raheem Sterling for the tap-in at the far post.
