The Gunners threw Granit Xhaka back into the line-up after his suspension at the expense of Albert Sambi Lokonga while Spurs recalled Lucas Moura, Japhet Tanganga and Davinson Sanchez.
Mikel Arteta's charges had a perfect start after Emile Smith Rowe opened the scoring in the 12th minute with a close-range finish inside the box thanks to a cut-back by Bukayo Saka.
It got better for the home side as Pierre-Emerick Aubameyang doubled their lead just before the half-hour mark from almost a similar move on the left side, although he finished off a pass from Smith Rowe.
Arsenal were oozing confidence from then on, with Saka giving his side a three-goal lead ten minutes before the half-time break with a low strike inside the box from a solo-run, after a missed challenge by Harry Kane.
Kane came the closest to pulling a goal back for Nuno Espirito Santo's troops eight minutes before the half-time interval with a close range header at the near post that missed just wide.
Espirito Santo made a double change at half-time, bringing on Oliver Skipp in place of Dele Alli while Emerson Royal came on for Tanganga at the back.
Kane came close twice in a short space of time around the mark, as he forced a save out of shot-stopper Aaron Ramsdale and dragged a shot just wide, after getting behind the Arsenal defence on both occasions.
Son Heung-Min pulled a goal back for the visitors ten minutes later with a side-footed strike inside the box after a cut-back by Reguilon.
Moura came close to pulling a second goal back with a deflected shot that was dipping into goal but Ramsdale tipped his effort into the crossbar as Arsenal held on to claim a derby win that moved them up to tenth on the log, equal on points with 11th-placed Spurs.
