Spurs' European Championship stars were expected to return for pre-season this week, but Sky Sports reported that the England skipper did not show up at all.
Kane reportedly informed the North London outfit that he wants to leave to win trophies and get regular UEFA Champions League football.
Guardiola confirmed City's interest in last season's Premier League Golden Boot winner.
"Harry Kane is an exceptionally talented striker and of course, we're very interested," he said, as per the Sun.
"But if Tottenham don't want to sell… Of course, we'll try."
Spurs chairman Daniel Levy is reportedly holding out for £160million, but City are looking to negotiate if their league rivals are also keen to talk.
"If Tottenham don’t want to negotiate, it is finished.
"If they want to negotiate, many clubs would like to sign him. We are not an exception. We are very interested in him."
