Paulo Dybala and Gonzalo Higuain lead Juve's attack against their Serie A rivals, while the likes of Edin Dzeko and Diego Perotti were expected to make things happen for Roma.
Both sides started the match strongly, with chances coming along for both teams, but it was the Bianconeri who took the lead after 29 minutes.
Brazil left wingback Alex Sandro picked out former Bayern Munich striker Mario Mandzukic, who collected the ball in the box and rounded goalkeeper Alisson before firing into an empty net.
The score stayed 1-0 going into the break, after which plenty of changes were made by both sides, which saw the game lose some of its momentum.
Dybala thought he had scored five minutes after half-time, but his strike was ruled out as he was offside when Higuain picked him out with a clever flick.
Ex-Manchester City striker Dzeko then fired Roma back into the game in the 74th minute, tapping in a great cross from Aleksandar Kolarov, his former teammate in England and new colleague in Italy.
The match ended 1-1 after 90 minutes, which meant that penalties would decide the clash, and it was Roma who were went first. Marco Tumminello put his side under pressure as he missed Roma's first penalty, which turned out to be the only miss in the shootout.
With the scores tied at 4-4, new signing Costa stepped up for Juve and his low effort gave the Bianconeri the win.
